Toxin profile, pigment composition, and large subunit rDNA phylogenetic analysis of an Alexandrium minutum (Dinophyceae) strain isolated from the Fleet Lagoon, United Kingdom

Paralytic shellfish toxins of a clonal culture of Alexandrium minutum isolated from the coastal Fleet lagoon, Dorset, UK in 2000 were analysed using high performance liquid chromatography with post column derivatisation and fluorescence detection. This analysis revealed a unique toxin composition profile showing the presence of gonyautoxin-3 (GTX-3; 54.3 mole%), gonyautoxin-2 (GTX-2; 9.6 mole%) and saxitoxin (STX; 36.1 mole%). The average toxicity of the culture was 3.8 pg STX-equiv. cell-1. Total toxin content varied from 5.6 fmol cell-1 on day 1 to a maximum of 16.8 fmol cell-1 during early stationary phase. In the UK, Paralytic Shellfish Poisoning (PSP) has been previously associated with the species Alexandrium tamarense. Blooms of A. tamarense have previously been described in the Fleet lagoon, but PSP toxins have not been detected in locally cultivated oysters. A. minutum has not previously been reported to occur at this location, however, the shallow lagoon may provide a favourable environment for this species to bloom and the identification of a toxic strain indicates the potential for future shellfish contamination. This study has established that A. minutum from the Fleet lagoon contains saxitoxin and related biotoxins and may contribute to PSP events in the locality.
